UPF1 promotes the formation of R loops to stimulate DNA double-strand break repair

چکیده

Abstract DNA-RNA hybrid structures have been detected at the vicinity of DNA double-strand breaks (DSBs) occurring within transcriptional active regions genome. The induction hybrids strongly affects repair these DSBs, but nature and how they are formed remain poorly understood. Here we provide evidence that R loops, three-stranded containing displaced single-stranded (ssDNA) can form sub-telomeric DSBs. These loops generated independently resection induced alongside two-stranded on ssDNA by resection. We further identified UPF1, an RNA/DNA helicase, as a crucial factor drives formation to stimulate resection, homologous recombination, microhomology-mediated end joining damage checkpoint activation. Our data show actively DSBs facilitate repair.

DNA double-strand break repair

The integrity of genomic DNA is crucial for its function. And yet, DNA in living cells is inherently unstable. It is subject to mechanical stress and to many types of chemical modification that may lead to breaks in one or both strands of the double helix. Within the cell, reactive oxygen species generated by normal respiratory metabolism can cause double-strand breaks, as can stalled DNA repli...

BMI1-mediated histone ubiquitylation promotes DNA double-strand break repair

Polycomb group (PcG) proteins are major determinants of cell identity, stem cell pluripotency, and epigenetic gene silencing during development. The polycomb repressive complex 1, which contains BMI1, RING1, and RING2, functions as an E3-ubuiquitin ligase. We found that BMI1 and RING2 are recruited to sites of DNA double-strand breaks (DSBs) where they contribute to the ubiquitylation of γ-H2AX...

Double strand break repair.

DNA double-strand breaks (DSBs) are the most dangerous form of DNA damage and can lead to death, mutation, or malignant transformation. Mammalian cells use three major pathways to repair DSBs: homologous recombination (HR), classical nonhomologous end joining (C-NHEJ), and alternative end joining (A-NHEJ). Cells choose among the pathways by interactions of the pathways with CtIP and 53BP1. HR i...
